Instrumentation

Learn about the instrumentation provided by the Pharmacoanalytic Shared Resource (PhASR) to OSUCCC – James cancer researchers.

Place an Order in eRAMP

The PhASR supports pre-clinical and clinical research involving drug development or evaluation of chemopreventive and carcinogenic properties of foods, plants, micro-organisms, and other environmental constituents. This is achieved by providing high-quality, cost-effective, and reliable bioanalytical method development and quantitative sample analysis utilizing bioanalytical systems ideally suited for quantitation of small molecule drugs, metabolites, and endogenous large and small biomolecules.

Primary PhASR instrumentation comprises four liquid chromatography/mass spectrometry (LC-MS/MS) systems and one multimode plate-reader/plate washer system.

Liquid Chromatography/Mass Spectrometry System Details

Sciex ZenoTOF 7600: A high resolution QTOF mass spectrometer equipped with TwinSpray TurboV Source and coupled to an Agilent 1290 Infinity II high speed pump, multicolumn thermostat, and temperature controlled multisampler.

ThermoScientific TSQ Altis: A triple-quadrupole mass spectrometer equipped with heated electrospray ionization probe and coupled to a Thermo Vanquish ultra-high pressure liquid chromatography system (UHPLC, degasser, quaternary pumps, heated column compartment, autosampler).

ThermoScientific TSQ Quantiva: A triple-quadrupole mass spectrometer with ultra-high-sensitivity (APCI and heated ESI ionization probes) coupled with a Thermo Vanquish ultra-high pressure liquid chromatography system (UHPLC, degasser, quaternary pumps, heated column compartment, autosampler). This system also incorporates an Easy-Spray nano-flow electrospray ionization source to achieve the maximal sensitivity possible for accurate quantification of small molecules, peptides, or other biomolecules in complex biological matrices or in small sample quantities.

ThermoFinnigan TSQ Quantum Ultra: A triple-quadrupole mass spectrometer with extended mass range and heated electrospray ionization probe connected to a Dionex RSLCnano system (with ternary loading pumps, quaternary nano-flow pumps, degasser, temperature-controlled autosampler and column compartment). ThermoScientific TSQ Altis: A triple-quadrupole mass spectrometer equipped with heated electrospray ionization probe and connected to a Thermo Vanquish ultra-high pressure liquid chromatography system (UHPLC, degasser, quaternary pumps, heated column compartment, autosampler).

Multi-Mode Plate-Reader Platform

BioTek Synergy™ LX: A multi-mode plate-reader capable of detection using monochromator-based absorbance from 200 nm to 999 nm, filter-based top fluorescence, and luminescence. This system has linear, orbital, and double-orbital shaking, and utilizes Gen5 software.

BioTek 50™ TS Washer: A compact, versatile plate washer for ELISA, cell-based and bead-based assays. This plate washer has buffer switching and biomagnetic separation capabilities.

The PhASR also maintains full access to other resources necessary to develop and run biological correlative assays, including UV-Vis and fluorescence spectrometers, plate-readers, centrifuges, and freezer and refrigerator storage.

WinNonlin, NONMEM and SimCYP Pharmacokinetic Analysis Software Packages are also available for data analysis.
